Default Dyno tuning

Just two questions.
1. What gear do they do it in?
I have a t5 and some dudes say they do it in 2nd others do it in 4th. whats the deal ?
2. Also what juice should i have in the tank ?
I know the high octane **** will give...hopefully better output but i dont always use it, actually i generally run on regular.
Umm i think thats it.
Cheers fellas

I like to run on the same fuel as i would use on a daily basis, I might run a premium fuel if I want to try get a better figure.

Dyno runs should be done in the gear that gives a 1:1 ratio, most auto's are in 3rd for this reason.
